Display Method:
A whole blood microculture method for obtaining metaphase chromosome and a technique of Trypsin-Giemsa banding were used in this study.T-G banding chromosomes of man are compared with those of golden monkey and rhesus monkey.Three cells of each species were analysed.The results show that the banding pattern of each pair of homologous chromosomes in different cells of the same species is analogy,but for cells in different species the banding pattern of most chromosomes is partly similar and that of only a few chromosomes is almost completely analogy.
The azo dye method for acid phosphatase and "Gomori" method for 5 Nucleotidase were used to identify the T,and B lymphocytes of mouse lymph node and thymus cytochemically.By employing these methods,the estimation of ACP and 5 N activities of parallel samples from the lymph node and thymus cell suspensions were carried out in order to verify the ACP and 5N positive lymphocyte percentages obtained previously.The ACP and 5 N positive lymphocytes thus estimated for lymph node were found to be 67% and 31%,whereas those for the thymus were around 97% and 10% respectively.Above mentioned results were not only well fitted with each other but also in good accordance with those already published.
The crude venom of scorpion Buthus martensii Karshi (from Liaoning) was fractionated into 12 protein peaks on CM-Sephadex C-50,Two toxins were purified from the eighth fracton by rechromatography on CM-Sephadex C-50 and gel filtration on Sehadex G-50.Their homogeneties were demonstrated by polyacrylamide gel electrophoresis,isoelectric focusing electrophoresis and SDS-polyacrylamide gel slab electrophoresis.The molecular weight of the toxins,calculated from measurement in SDS-Discontinuous polyacrylamide gel slab electrophoresis,are 8,980 and 8,660 respectively.Their isoelectric points are 7.58 and 7.90 respectivey by isoelectric focusing electrophoresis.The amion acid composition of the toxin Ⅰ was determined.The crude venom showed hyaluronidase activity but on phospholipase A[2],α-amyase,arginine esterase,acetylcholinesterase,phosphomonoesterase,L-amino acid oxidase as well as proteinase activity.The LD[50] of crude venom of scorpion Buthus martensii Karshi was found to be 10.3 μg/g for mice.The protein of molecular weight less than 10,000 dalton could be determined by SDS-disc polyacrylamide gel slab electrophoresis (13%).The results obtained were more satisfactory.
