Abstract
    ThecompletemitochondrialgenomesequenceofPielomastaxzhengiwasdeterminedbyusinglongPCRandconservedprimerwalkingapproaches.TheresultsshowedthattheentiremitochondrialgenomeofPielomastaxzhengiis15602bplongwithA+Tcontentof71.8%.All37genesareconservedinthepositionsobservedinthoseofLocustamigratoria.Allthegenesarecloselyassembledbyleaving10intergenicspacersinbetween.Thoseintergenicspacersare47bpintotal(excludingtheA+Trichregion),withindividualsizerangesfrom1bpto20bp.Inaddition,thereareatotalof52bpoverlappingsequencesamong14genes,rangingfrom1−8bp.Allprotein-codinggenesstartwithatypicalinitiationcodonininsects,ATN.Twelveprotein-codinggenesusetheusualTAAandTAGterminationcodons,whereas,theND5geneshaveanincompleteterminationcodon(T).ExceptthetRNASer(AGN),whoseDHUarmisabsent;alltheother21tRNAgeneshavetypicalclover-leafsecondarystructures.ButinPielomastaxzhengi,thesecondarystructuresoffivetRNA(tRNACys,tRNALys,tRNAPhe,tRNAPro,tRNAArg)genescannotbepredictedbytheconventionalmethodsastheyhaveonly3−4,ratherthan5basepairsintheTψCarm,while,thetRNALysandtRNAArghaveonly4basepairsintheanticodearm.ThepredictedsecondarystructuresoflrRNAandsrRNAhave6domainswith44helicesand3domainswith30helices,respectively.TheresultsoftherRNAsecondarystructurecomparisonshowedthatPielomastaxzhengiismorecloselyrelatedwithLocustamigratoriatibetensisthanRuspoliadubia.Likemost&
