基于形态学和线粒体COI基因的有孔团水虱分类鉴定

Identification of Sphaeroma terebrans via morphology and the mitochondrial cytochrome c oxidase subunit I (COI) gene

  • 摘要: 有孔团水虱(Sphaeroma terebrans)为甲壳等足类蛀木动物。有孔团水虱广泛分布于世界热带和亚热带红树林。以往有孔团水虱的鉴定通常基于形态特征,缺乏分子方面的研究。尾节外肢上的齿数和第七步足长节的长度是影响有孔团水虱鉴定的主要形态特征。本研究通过形态学特征和分子数据对有孔团水虱进行鉴定,并验证了以线粒体COI基因作为DNA条形码鉴定团水虱属的有效性。线粒体COI基因序列分析显示,有孔团水虱、光背团水虱(Sphaeroma retrolaeve)和团水虱Sphaeroma serratum的种间距离大于种内距离,并且两者之间无重叠区域;基因遗传距离和核苷酸多样性(π)在有孔团水虱群体中没有差异。研究表明,线粒体COI基因可以作为有效的DNA条形码鉴定团水虱属,而尾节外肢上的齿数和第七步足长节的长度不能作为有孔团水虱可靠的分类特征。

     

    Abstract: Sphaeroma terebrans, a wood-boring isopoda, is distributed worldwide in tropical and subtropical mangroves. The taxonomy of S. terebrans is usually based on morphological characteristics, with its molecular identification still poorly understood. The number of teeth on the uropodal exopod and the length of the propodus of the seventh pereopod are considered as the major morphological characteristics in S. terebrans, which can cause difficulty in regards to accurate identification. In this study, we identified S. terebrans via molecular and morphological data. Furthermore, the validity of the mitochondrial cytochrome c oxidase subunit I (COI) gene as a DNA barcode for the identification of genus Sphaeroma, including species S. terebrans, S. retrolaeve, and S. serratum, was examined. The mitochondrial COI gene sequences of all specimens were sequenced and analysed. The interspecific Kimura 2-parameter distances were higher than intraspecific distances and no intraspecific-interspecific distance overlaps were observed. In addition, genetic distance and nucleotide diversity (π) exhibited no differences within S. terebrans. Our results revealed that the mitochondrial COI gene can serve as a valid DNA barcode for the identification of S. terebrans. Furthermore, the number of teeth on the uropodal exopod and the length of the propodus of the seventh pereopod were found to be unreliable taxonomic characteristics for S. terebrans.
